About the author.Optimization of Na+ transport in biotechnologically important Halobacillus species by a combination of screening and random mutation approaches. Mutations are known to improve salt tolerance in many microorganisms. In the present work, the effect of random mutagenesis on the salt tolerance of two different Halobacillus species, H. halobius and H. hispanicus, was investigated. The genes encoding the sole Na(+)/H(+) antiporter of the respective organisms, AhnK1, were sequenced, and the homologous expression of the protein was confirmed by Northern blot analysis and Western blot. Random mutagenesis was performed by error-prone PCR (epPCR) on the HhK1 gene. This resulted in 10 mutant libraries for each species, which were screened for enhanced salt tolerance. The effects of Na(+) deficiency on cell growth and expression of the Na(+)/H(+) antiporter were determined by monitoring the colony-forming ability of the HhK1 mutators after exposure to Na(+)-deficient medium.
